Markers for Hematopoietic Stem Cells

We have a long-standing program to define new markers for hematopoietic stem cells (HSC). Previously, we showed that HSC express Thy-1 but lack markers associated with mature cells (Thy-1loLin-) [Cell, 1986; 44:653. We also demonstrated expression of CD4 on HSC [Blood, 1992; 80:1717] a finding that has implications for the understanding of the pathogenesis of AIDS. To define new markers on HSC, an extensive panel of monoclonal antibodies was generated against Lin- cells and many of these detect new markers on HSC. One of these mAb, named Fall-3 [J Exp Med, 1991; 174 : 161] separates HSC from the earliest lymphoid progenitors. A major focus in the lab is to characterize additional HSC markers and in particular, to define markers specific for Linage-biased HSC.
